TB-500 (TB500 or Thymosin β-4 Active Fragment) is a synthetic heptapeptide corresponding to the N-terminal actin-binding domain of endogenous Thymosin β-4. It modulates cytoskeletal dynamics by sequestering G-actin monomers and promoting localized F-actin polymerization, thereby enhancing cellular motility and migration. TB-500 upregulates angiogenic growth factors (e.g., VEGF, FGF-2) via MAPK/ERK and PI3K/Akt signaling, driving neovascularization in ischemic or injured tissues. Concurrently, it attenuates pro-inflammatory cytokines (TNF-α, IL-6) through NF-κB inhibition, creating a reparative microenvironment. In in vitro and rodent models, these combined actions support accelerated tissue regeneration, angiogenesis, and reduced inflammatory damage.
